薄层色谱法的优缺点及操作步骤

①优点:操作方便、设备简单、显色容易等特点,同时展开速率快,一般仅需15~20min;混合物易分离,分辨力一般比以往的纸层析高10~100倍,它既适用于只有
0.01μg的样品分离,又能分离大于500mg的样品作制备用,而且还可以使用如浓硫酸、浓盐酸之类的腐蚀性显色剂。

(3)CAOZUOBUZHOU

①薄层板制备:除另有规定外,将1份固定相和3份水在研钵中向一方向研磨混合,去除表面的气泡后,倒入涂布器中,在玻板上平稳地移动涂布器进行涂布(厚度为
0.2~O.3mm),取下涂好薄层的玻板,置水平台上于室温下晾干,后在110℃烘30min,即置有干燥剂的干燥箱中备用。使用前检查其均匀度(可通过透射光和反射光检视)。

④显色:取出的薄层板,立即喷洒显色剂,使其显色,计算Rf值或使之出现各种有色的斑点,与标准进行对照比较,可确定其成分。
